FOXO4-DRI 10mg/10vials is a sterile lyophilized research peptide. FOXO4-DRI was developed by the Peter de Keizer lab and published in Cell (2017) as a tool peptide for the selective elimination of senescent cells. The molecule is a D-retro-inverso (DRI) peptide — synthesized with D-amino acids in reverse sequence — derived from the interaction interface between FOXO4 and p53, designed to disrupt that interaction selectively in senescent cells.

Mechanism
In senescent cells, FOXO4 sequesters p53 away from the mitochondria and prevents apoptosis, which is part of why senescent cells accumulate and persist. FOXO4-DRI is designed to competitively disrupt the FOXO4-p53 interaction, releasing p53 to translocate to the mitochondria where it triggers apoptosis specifically in cells that have built up senescent stress. The DRI configuration confers proteolytic stability while preserving the binding-interface geometry of the parent sequence.
In the original publication, FOXO4-DRI was shown to selectively kill senescent cells in vitro and to rescue several aging-associated phenotypes in mouse models without affecting healthy cells. It has since become one of the standard tool peptides in preclinical senolytic research.
Typical research dose range
Published preclinical senolytic protocols using FOXO4-DRI in mouse models have typically used intraperitoneal administration at quantities in the range of 5 mg/kg every other day across multi-week cycles. Researchers should consult the original Cell 2017 paper and follow-on senolytic literature when designing protocols.

FOXO4-DRI is one of the canonical tool peptides in preclinical senolytic and longevity research. It is frequently studied alongside Epithalon (telomerase-activity work), NAD+ (cellular bioenergetics), and SS-31 (mitochondrial oxidative stress) in multi-arm aging-axis study designs.
Reconstitution and storage
Reconstitute each vial with 1-2 mL of bacteriostatic water for the target working concentration. Use sterile technique. Once reconstituted, store at 2-8 °C and use within 30 days for best stability.
Lyophilized vials should be kept refrigerated at 2-8 °C until first use. For long-term storage beyond six months, store at -20 °C in the original sealed vial. Avoid repeated freeze-thaw cycles.
